A membrane component involved in the transport of adenosine in adipocytes has been identified utilizing the techniques of photoaffinity labeling with the adenosine derivative, 8-azidoadenosine. In the absence of light, adenosine and 8-azidoadenosine exhibited similar transport characteristics. In addition, adenosine was shown to be a competitive inhibitor of 8-azidoadenosine uptake, and the photoprobe, a competitive inhibitor of adenosine uptake. Analysis of the nucleotide metabolites indicated that the photoprobe was metabolized in a similar fashion to that observed for adenosine. Several nucleoside transport inhibitors were also equally effective in inhibiting the uptake of both nucleosides. These results suggest that 8-azidoadenosine is transported by the same membrane system as adenosine. Photolysis of 8-azido[2-3H]adenosine in the presence of adipocytes resulted in the covalent incorporation of the photoprobe into the plasma membrane fraction. Analysis by sodium dodecyl sulfate-polyacrylamide gel electrophoresis indicated that essentially all of the radioactivity was incorporated into a glycoprotein with a molecular weight of 56,000. This labeling was inhibited by greater than 90% when the photolysis was carried out in the presence of excess adenosine or the transport inhibitors, persantin or theophylline. Fractionation of the labeled plasma membranes by dialysis against water (pH 9.5) indicated that approximately 75% of the radioactivity was associated with a glycoprotein which resisted solubilization by this procedure. These results suggest that the major labeled species is a 56,000 Mr intrinsic membrane glycoprotein which may function as a component of a transmembrane assembly involved in the transport of adenosine. 相似文献

Abstract Climatic data and collection records for the cactophilic Drosophila aldrichi and Drosophila buzzatii for 97 localities were used to examine the effects of geographical location, season, host plant species and climatic factors on their range and relative abundance. Temporal variation in relative abundance was assessed from monthly collections over 4 years at one locality. Effects of weather variables over the 28 days before each collection were examined. A generalized linear model of the spatial data showed significant geographical variation in relative abundance, and significant climatic effects, with the proportion of D. aldrichi higher in the warm season, and increasing as temperature variation decreased and moisture indices increased. The temporal data gave generally concordant results, as D. aldrichi proportion was higher in summer and autumn, and increased as maximum and minimum temperatures increased, and as variation in maximum temperature decreased. In a laboratory competition experiment, D. aldrichi eliminated D. buzzatii at 31°C, but was itself eliminated at 18°C and 25°C. The range of D. buzzatii is constrained only by availability of its host plant, Opuntia species, although its relative abundance is reduced in the northern part of its distribution. The range of D. aldrichi, from central Queensland to northern NSW, Australia, is entirely within that of D. buzzatii, and its relative abundance decreases from north to south. Both climate and weather, particularly temperature variability, have direct effects on the relative abundances of the two species, and both likely act indirectly by influencing the outcome of interspecific competition. 相似文献

An earlier field study on the fifteen-spined stickleback (Spinachia
spinachia) showed that frequent malemale interactionsresult in high frequencies of sneaking and egg stealing. Moreover,sneaking behavior was performed not only by males adoptingalternative mating strategies, but also by males with theirown nests. The advantage of sneaking is easily understood,
but it is more difficult to explain the evolutionary benefitof stealing eggs from other males. I investigated whether malessuffering from sneaking adjust their paternal effort in relationto their degree of paternity. I also examined whether femalesprefer males that have more eggs in their nests, as this couldexplain egg stealing. There was no relationship between thedegree of paternity and fanning activity, hatching success,or nest defense. However, the older the eggs become, the morethe males increase their attack rate toward potential egg predators(goldsinny wrasse and shore crabs). Thus, males adjusted theirlevel of defense to the amount of energy and time already investedin the clutch. Females did not prefer males with more eggs intheir nests.
